报告题目:Sulfoxide Polymers as Nanomedicines

Polymer-based therapeutics are increasingly being used for the treatment of various diseases, including cancer. However, their rapid clearance from the body and immunogenicity often hinder successful clinical outcomes. The formation of conjugates with poly(ethylene glycol) (PEG) is the most commonly used strategy to improve the in vivo pharmacokinetic profile of proteins by providing "stealth" properties. Despite the desirable outcomes offered by PEG, the immunogenicity and hypersensitivity that are found to be associated with PEG have generated concerns, prompting the development of alternative polymers for protein conjugation. In response we have developed a series of polymers containing the highly hydrophilic sulfinyl moiety, for example poly(2-(methylsulfinyl)ethyl acrylate, PMSEA. Initially we found that oxidation of thioethers produced a change in solubility and that this could be exploited as a responsive MRI agent. On recognizing the potential of this class of polymer for imparting high levels of hydration to materials, we demonstrated the use of PMSEA and derivatives as PEG alternatives in nanomedicines. More recently grafting of sulfinyl polymers onto substrates has been shown to dramatically reduce protein adhesion and cell attachment, and applications in medical devices are being explored.

报告人简介(中文):Prof. Andrew Whittaker is Senior Group Leader within the Australian Institute for Bioengineering and Nanotechnology, at The University of Queensland (UQ). He graduated from UQ with a Ph.D. in 1987, held postdoctoral positions at the University of Science and Technology Languedoc, Montpellier and the Max-Planck Institute for Polymer Research, Mainz, before working for three years at BP Research in Sunbury-on-Thames, U.K. He returned in UQ in 1991, and has developed a large program of work based on polymeric nanoscience. He has been the recipient of many awards, notably in 2015 the Chinese Academy of Sciences President’s International Fellowship, and, on three occasions, High-End Foreign Expert of the PRC. Whittaker has held many leadership positions, and most recently is the immediate Past-President of the Pacific Polymer Federation.
